After Creating a new user in SugarCRM, In the Access Tab we assign the User a Role. How do we do that using Rest API. which Endpoint helps here ?

I think this is a bit tricky to try to do via API. Users get related to ACLRoles via the aclroles relationship.
If you know the right ACLRole ID, then you could try something like:
POST /Users/:record/link/aclroles/:remote_id
