The issue may be caused by a database timeout. To determine if this is the cause, go to Admin > System Settings and enable 'Log slow queries'. Also, ensure the 'Slow query time threshold' is set to a reasonable number (the default is 5000 msec). You could set this to 2000 or 3000, but based on the behavior you are seeing 5000 should be restrictive enough if this is a database issue.
Then, reproduce the issue and investigate the sugarcrm.log file to see if a slow query was reported. If you have direct database access, run an EXPLAIN on that query to identify where the performance bottlenecks are occurring. Depending on the results, adding an index on the one or more fields may remedy the issue. If you are unsure of how to proceed, please provide the EXPLAIN results.
If Sugar does not log any slow queries when encountering the search timeout, I recommend checking the server/PHP error log(s) for any errors corresponding with the action.

My plan is to create index using database tool and also to create custom index for the custom field . what is the purpose of defining the custom index in a Vardefs Extension ?
Is it any relation with the elastic search and custom index
If you create a vardef index on a custom field, the only purpose that serves will be to allow users to leverage that index as a duplicate record flag on import. It does not have an impact on database indices or any other function. If you create a vardef index on stock fields, it has the dual purpose of creating the duplicate record check as well as a database index.

Please check the elastic search once as well. It could be that the elastic search is down. That sometimes gives a 500 internal server error as well.

If elastic search down it will affect the whole searching,But in my it was only happen when I am trying to search with a newly added custom field.
I tried by adding index on the _cstm table but still showing the issue. Can you suggest any other ways to solve this issue ?
